I agree with Dr. Humphries that most of the time this is normal pigmentation of the iris and not a concern. It sounds like you said they are growing and changing so this could be a concern. You should have your veterinarian examine him closely. He/she may even refer you to an animal ophthalmologist. That is what I usually do with any that look suspicious to me.
You can have melanoma in the eye without any other symptoms. It can spread, so if it is likely melanoma, it is better to have the eye removed before it spreads. They do great with the surgery and afterwards adapt very well.

Just guessing I would think you are seeing normal pigmentation of the iris. This is rather common. I cannot say for sure, but most likely not a melanoma. This is especially true when you consider you have watched this for a couple of years now and it has not grown.
At 6 years it is time for a good physical exam anyway, have your veterinarian look closely at those spots.
