It depends on the woman.  Plan B didn't affect me at all.  My period came right on time.  It can make some women bleed long before they're due.  It can make some women bleed later than when they're due.  It all depends on the person.  I'd say if it's more than a couple weeks late, have her take a test.

However, don't sit there stressing about it.  The likelihood of her being pregnant from this is so very unlikely, and stress can delay a period (as can if she starts going on an extreme exercise regimen or gets sick).

Pre-ejaculate can contain a small amount of sperm, but as I said, it is unlikely, especially if you gave her the pill right after.  The sooner emergency contraception is given, the more effective it is going to be.

It's unlikely, but Plan B should only be used in emergency situations, not all the time.  She needs to get on a regular regimen of birth control and/or you need to use a condom to avoid pregnancy.  (Note: Most birth control pills require a month of consistent, proper use before it will be effective.)  It is not good for her body to have Plan B or any other emergency contraception all the time.
