What they mean is first thing in the morning after you have slept.  6 am would be better than 12 midnight.  The reason why they say first mornings urine is best to use is because the concentration of hormones in the urine is greater and it is easier for the test to pick up the hormone. If you are testing right at when you missed your period, this is better because the concentration is such that you may need the most in your urine to get a positive result as opposed to being say 6 weeks pregnant when you can test any time of day and get a positive because there is so much of the hormone.  Does that make sense?  So, they want you to sleep all night, then test when you wake up.  good luck

You can always test at any time of day.
The first morning pee will have a bigger 'concentrated' amount of the particular hormone the test is looking for because you haven't peed all night, that's why most people say to use the first morning pee.
